AbstractWe have examined the ability of cationic liposomes to mediate the transient transfection of human monocyte and lymphocyte cell lines, as well as of peripheral blood monocytes (PBM) freshly isolated from normal human donors. We found that cationic liposomes can mediate high level expression of expression vectors coding for either an intracellular (chloramphenicol acetyltransferase) or a secreted (interleukin-2) gene product in the monocyte and lymphocyte cell lines. However, the human PBM could not be transfected using this approach. The use of a T lymphocyte-specific promoter element (IL-2) appeared to restrict cationic liposome-mediated transgene expression to lymphocytes, suggesting that transgene expression may targeted to specific cell types by the use of tissue-specific promoter elements.
