With the emergence of severe acute respiratory syndrome coronavirus 2, a new class of vaccines, BNT162b2 and mRNA-1273, were used for immunization. Both vaccines are based on an mRNA molecule that encodes the viral spike (S) glycoprotein of severe acute respiratory syndrome coronavirus 2. While efficacious at preventing severe manifestations of COVID-19 (coronavirus disease 2019), adverse effects have been reported, including hematologic disorders and cardiovascular complications such as myocarditis and pericarditis. In this context, a better understanding of the physiological responses to COVID-19-based mRNA vaccines and their short-and long-term effects on health are crucially needed.The vascular endothelium plays a central role in the pathogenesis of COVID-19 and may therefore be involved in some of the acute cardiovascular side effects of COVID-19 mRNA vaccines. However, only 2 studies have directly examined the impact of these vaccines on vascular function, and the assessments were conducted during different period following vaccination. 1, 2 In the short term, flow-mediated dilation of the brachial artery was transiently impaired in healthy adults 24 hours after a second dose of BNT162b2. 1 To date, it is still unknown whether mRNA-1273 vaccine elicits the same effect on the vasculature nor the potential mechanisms responsible for the transient changes in vasodilatory function postvaccination. The aim of this study was therefore to evaluate the short-term effects of COVID-19 mRNA vaccines (BNT162b2 and mRNA-1273) on vascular endothelial function and nitric oxide (NO)-mediated vasodilation in healthy adults 48 hours after vaccination.
