The possibility that the synthesis of late adenovirus serotype 2 proteins was regulated at the level of translation was examined. Comparison of the rate of synthesis of individual viral proteins with the activities of their messenger RNAs measuredin vitrosuggested that the translation of polypeptide IV and IX mRNAs might be specifically suppressedin vivo. However, comparison of the relative molar rate of synthesis of viral proteinsin vivowith the relative molar abundance of their mRNAs indicates that all viral mRNAs studied which possess the late three segment leader (II, 100K, III, IIIa, IV, V, pVI, pVIII and pVII) are translated with equal efficiencyin vivo. Polypeptide IX mRNA, however, is translatedin vivoapproximately five times as efficiently as the other late mRNAs. Efficient translation of this mRNA may be due to an extensive complementary region between the 5′-terminus of IX mRNA and the 3′-terminus of 18 Sribosomal RNA, or to the relative proximity of the 7mG cap to the initiating AUG codon.
