项目摘要

女性ホルモンの顕著な変動は、多くの女性に月経前症候群(PMS）、産後うつ、更年期障害、記憶障害などを引き起こす。女性ホルモンは、受容体のサブタイプの発現変化やシナプス数の変化などの神経形態変化や、受容体の感受性の変化などによる神経伝達効率の低下など神経機能を変化させ、多くの症状の誘因となることが考えられている。これらの詳細な神経形態・機能変化を解析するため、PMSの病態解明や治療薬の効果検討を目的としたPMSモデル細胞の作成を目指した。女性ホルモンのプロゲステロンは、脳内において神経ステロイドを合成し、GABA神経に作用する。神経ステロイドの中でも、3α,21-Dihydroxy-5α-pregnan-20-one(THDOC)はGABAA受容体調節物質であり、GABAA受容体が強く応答する。そこで、GABAA受容体調節物質であるTHDOCやプロゲステロンがGABA神経およびグルタミン酸神経に及ぼす影響についてアストロサイト・単一ニューロン共培養系を用いてPMS神経モデルとなりうるか形態学的解析により検討した。多くの漢方薬は女性ホルモン変動関連疾患に用いられているが、それぞれの作用機序の差異や、作用の中心を成す構成生薬や有効成分などの詳細は不明である。そこで当帰芍薬散、加味逍遙散、桂枝茯苓丸をはじめとする婦人科処方の漢方薬および構成生薬をアストロサイト・単一ニューロン共培養系に処置し、ニューロステロイド合成酵素（5a reductase）、神経栄養因子（NGF, BDNF）などニューロン形態関連因子の挙動を解析した。
