项目摘要

鼻腔炎では多様な分子免疫応答が引き起こされるが、免疫エフェクター細胞とIL-17等のサイトカイン標的細胞との相互作用機序を病態ごとに詳細分類できれば治療戦略に大きく貢献できると考えられる。浮遊細胞の培養上清中の分泌サイトカインをBioPlexサスペンションアレイ測定にて網羅的に解析し各病態ごとのプロファイリングによる分類評価を行う。当施設で所有するBioPlexサスペンションアレイは、培養上清中に分泌されたサイトカインを多項目同時に検出することができる安価で簡便なアッセイシステムである。同システムにより鼻ポリープ由来浮遊細を用いた効率的なサイトカイン発現プロファイリング法を検証する。線維芽細胞の増殖と活性化は炎症細胞や、その産生するケミカルメディエーターにより直接的あるいは間接的に影響を受けている。本研究では細胞増殖指示薬として簡便で効率的なWST-8を用い、IL-17を中心とした副鼻腔炎関連サイトカインに対する反応として細胞増殖能および増殖停止能の測定を検証した。線維芽細胞はサイトカインを放出し炎症反応をコントロールすることが知られている。本研究では無刺激の培養上清中の分泌サイトカインとIL-17等により惹起されるサイトカイン分泌パターンを上記のBioPlexアッセイ系にて比較分析することにより、各病態による細胞応答性の変化を解析した。上記解析により得られた各病態に対する表面抗原・細胞内タンパク質発現パターン、サイトカイン分泌パターン、線維芽細胞のサイトカイン応答性の結果を総合的に様々なクラス分け）を行い臨床データとともにデータベース化する。このデータベースをこれまでの我々の報告における病理データ・検体由来患者の臨床所見との比較検証を行う。このin vitro評価法にて同定された分子免疫機構を臨床データと照合した。
