项目摘要

動物性脂肪を多く含む不健康な食事の過剰摂取は、腸管バリアおよび血液脳関門の障害を引き起こし、脳における神経炎症を促進する。慢性的な神経炎症は認知機能を障害することが報告されている。健康長寿食として有名な地中海食の中核を成すエクストラバージンオリーブオイル(EVOO)は心血管系の保護効果のみならず、近年では認知機能の向上や腸内フローラのバランス改善効果が示唆されている。このような背景を踏まえ、本研究では老化促進型認知機能障害モデルマウス(SAMP8)にEVOOを与え、認知機能障害及び腸と脳のバリア機能障害を予防・改善できる可能性を検証した。Tight junction protein(TJP)は腸管上皮細胞において腸内細菌の体内への侵入を防ぎ、また血液脳関門においては末梢組織からの炎症誘発因子の脳内への侵入を防ぐ。この腸と脳に共通して発現するバリア蛋白質群の発現量の変化を評価した。16週齢のSAMP8マウス(N=10/群)に低脂肪食、45%ラード含有高脂肪食、35%EVOO含有高脂肪食を12週間与え、正常な老化プロファイルを示すSAMR1マウスをコントロール群として同様に給餌した。認知機能を評価するために、新規物認識試験、位置認識試験、Y字迷路試験を実施した。さらに、それぞれの群の半数のマウス(N=5/群)から、脳組織(海馬、前頭前野)、回腸、盲腸、結腸組織を採取し、TJPのmRNA及びタンパク質の発現レベルを解析した。EVOO給餌12週間後において、各群間で認知機能及びTJPの発現レベルには有意差を認めなかった。そこで、残りのマウス(N=5/群)は給餌期間を22週間延長し、再度Y字迷路試験を用いて認知機能を評価したが、各群間に有意差を認めなかった。その後、コロナ禍の影響による研究代表者の家庭事情の変化により、研究の継続が困難となり、辞退させていただくこととした。
