The purpose of this research is to identify and investigate the role of active constituents of seaweeds in obesity associated with SMS imbalance. Preliminary inhibition screening results showed Wakame as one of the potential candidates with 66% (SMS1) and 75% (SMS2) at 100 μg/mL. I have proposed the following objectives in the project(1) Perform solvent-fraction-based screening and identification of the active constituent in Wakame (2) Examine the in vitro effects of the active compound on lipid droplet accumulation and oxidation by cell-based assay. As proposed the wakame samples were purchased on a large scale and grind into powder and its crude methanol extract was prepared three times. As the methanolic extract of wakame showed a promising result against SMS inhibition, the next step is to identify and investigate the role of an active constituent of wakame. further, solvent-based partitions were made. Solvents such as Hexane, Ether, Ethanol, butanol, and water-based screening have been performed and found that hexane fraction has potent inhibition activity. I am aiming to purify the active compound. Meantime other cell-related experiments are started, as soon as the compound is purified. This turn will help the development of wakame-based nutraceuticals to control obesity.
