本研究では、2010年1月以降に山口大学医学附属病院小児科で急性リンパ性白血病と診断し、維持療法で6-メルカプトプリンを投与した症例を対象とした。方法としては、患者末梢血中に含まれる正常白血球からキットを用いてDNAを抽出した。次に、キットを用いてPCRを行い、NUDT15 エクソン1およびエクソン 3領域を増幅した。続いて、増幅したPCR産物の塩基配列をダイレクトシーケンス法によりNUDT15多型の有無について解析した。ダイレクトシーケンスの結果、対象のうち21例が解析でき、16例がNUDT15多型なし、5例が多型ありであった。多型ありの内、3例がc.415C>Tのヘテロ接合性多型、1例がc.52G>Aとc.415C>Tの2つのヘテロ接合性多型、残りの1例がc.415C>Tのホモ接合性多型とc.36_37insGGAGTCのヘテロ接合性多型を有していた。2つのヘテロ接合性多型を持つ症例について、ディプロタイプ解析のためクローニングを用いた解析を行った。まず、正常白血球から抽出したmRNAを用いて相補的DNA（cDNA）を合成した。合成したcDNAをキットを用いてクローニングし、合計9個のコロニーが得られた。ダイレクトシーケンスを用いて解析を行った結果、2種類の多型はそれぞれ別のアレルに存在することが分かり、複合ヘテロ接合性多型であるとわかった。また、同症例をデジタルPCRを用いてディプロタイプ解析を行った。まず多型を有するアレルが標識されるようなプローブを設計し解析した結果、c.52G>Aの多型を有する集団とc.415C>Tの多型を有する集団とに分かれ、デジタルPCRで複合ヘテロ接合性多型であることが確認できた。
