ユビキチンは、選択的細胞内蛋白分解を誘起する因子である。すなわち、“遊離型ユビキチン"が酵素群の作用で標的蛋白質に結合し、“マルチユビキチン鎖"を形成することで26Sプロテアソームによる標的蛋白質の分解を惹起する。本研究は、ラット肝癌におけるユビキチン動態の解析を目的として行った。ラットを0.06%3'-methyl-diaminobenzidine(3'-Me-DAB)含有固形試料を用いて140日間飼育し、肝臓癌を誘発した。この肝臓の癌部と非癌部及び正常ラット肝臓の各々から、トリス緩衝液に可溶な易様性画分と同緩衝液に不溶で8M尿素で可溶化される難溶性画分を調製した。各画分の単位蛋白あたりの“遊離型ユビキチン"と“マルチユビキチン鎖"を2種類の特異的イムノアッセイで定量し、以下のような知見を得た。1.すべての組織において、殆どの遊離型ユビキチンは易溶性画分にのみ認められた。一方、マルチユビキチン鎖は、易溶性および難溶性の両画分に見出された。2.癌部の遊離型ユビキチン量は、非癌部および正常の肝臓と比較し、有意に多かった。3.癌部の易溶性マルチユビキチン鎖量は、非癌部および正常肝臓よりも有意に多かった。4.癌部の難溶性マルチユビキチン鎖量は、非癌部および正常肝臓よりも多い傾向を示した。また、同様の肝癌ラットの血清マルチユビキチン鎖濃度は、正常ラットよりも有意に高値を示した。以上の結果から、肝癌ではユビキチン全体量が増加し、それが血清マルチユビキチン鎖濃度に反映されることが示唆された。今回の結果がユビキチン依存性蛋白分解の亢進を意味するのか、また、標的蛋白質は何かといった問題が、肝癌におけるユビキチン系の意義を解明する上で重要な今後の課題である。
