c-erbB-2/neu遺伝子をtransfectionさせたPC12培養細胞は、NGFなしで分化するには至らなかった。そこで、transfection、させた細胞にNGFを添加し、neuron様に分化させ、培養液中に放出された蛋白質を濃縮した。この濃縮した蛋白質をSDS-PAGEで展開してc-erbB-2/neu遺伝子をtransfectionさせなかった分化PC12培養細胞と比較したが、c-erbB-2/neu遺伝子由来の蛋白質を確認、他には目立った蛋白の新規発現は見つけられなかった。PC12培養細胞膜上には僅かながらERが存在する。PC12培養細胞がneuron様に分化する際、ERを高発現させるので、培養液中にestradiolを添加しておいた場合のERの出現傾向を、NGFで分化させたc-erbB-2/neu遺伝子をtransfectionさせていないPC12培養細胞とtrancfectionさせたPC12培養細胞について、RNase protection assay、及び免疫化学的染色で解析した。その結果、ER mRNA発現は、c-erbB-2/neu遺伝子をtransfectionさせたPC12培養細胞の方がtransfectionさせなかった分化PC12培養細胞に比べ、著明であった。
