项目摘要

1.放射線照射後のTc-99m MIBI集積変化 A549細胞では放射線照射後にTc-99m hexakis-2-methoxy-isbutyl-isonitrile[MIBI]集積が亢進した.その集積は照射直後-2時間と6-12時間後に2峰性の増加を示した.さらに放射線感受性の異なる2種のがん細胞株で検討したが,やはり放射線照射後にTc-99m MIBI集積が増加した.2.フローサイトメトリによるミトコンドリア膜電位の解析 フローサイトメトリによる3,3′-dihexyloxacarbocyanine[DiOC.(3)]を用いた解析から,A549細胞のミトコンドリア膜電位は照射直後および4-12時間後の2峰性の増大を示し,膜電位の変化がTc-99m MIBI集積の変化の主因であることが確認された.3.放射線照射後の細胞周期の変化とミトコンドリア膜電位の関連 フローサイトメトリによる解析からA549細胞は照射4-8時間後にG2/M期細胞が増加した(G2ブロック).上記のTc-99m MIBI集積の第2ピークはこの照射後のG2ブロックの時期とほぼ一致していたことから,細胞周期をカフェイン(照射後のG2ブロックを抑制することが知られている)を用いて変更し,ミトコンドリア膜電位への影響を検討した,その結果,G2ブロックが消失しても膜電位にはほとんど変化がみられず,膜電位は細胞周期に依存しないことが判明した.4.まとめ がん細胞は放射線照射後にTc-99m MIBI集積が増加し,集積の増加はミトコンドリア膜電位の増大による.膜電位の増大は細胞周期に依存しない.またミトコンドリア膜電位の増大は照射直後から生じることから,こめ膜電位の変化は照射後極めて早期に生じる細胞代謝上の反応と考えられた.研究成果の一部は平成12年11月に日本放射線腫瘍学会第13回学術大会にて発表した.
