「目的」本研究は水頭症進行時での脳細胞障害を明らかとするため先天性水頭症モデル動物の脳内Bcl-2の変動を基盤として即時型quantified RT-PCR法による検索を行なった。 「方法」従来から使用している久留米大学で見出された先天性水頭症モデル動物(LEW-HYR)の同腹で典型的な水頭症ラットと正常ラットを選択し,同週齢の正常群を対照として使用した。生後7および14日で断頭後、直ちに氷冷中で大脳皮質と小脳に分け,測定日まで-80℃で保存した。Bcl-2mRNAの測定は各脳部位をIsogenで組織混和しc-DNAを抽出後即時型quantified RT-PCR法で詳細な定量解析を行った。「結果」1)LEW-HYRの正常ならびに水頭症群におけるBcl-2mRNAの塩基配列は既に登録のあるラット脳内Bcl-2mRNAの配列と同一であることが遺伝子解析から明らかとなった.2)正常群において生後7日から14日にかけていずれの脳部位におけるBcl-2mRNAの発現は著明に低下した.3)典型的な水頭症群は生後7日目と14日目でのBcl-2mRNA発現に著明な変動は認められ無かった.しかし,いずれの脳部位,生後の日数における各値は,正常群と比較して明らかな発現の増加が認められた。しかも,この増加は水頭症の進行程度,重傷度により発現頻度の亢進が確認された。しかしながら,小脳においては,LEW-HYRの14日目以外では明確な変動が認められなかった。「考察」本研究の成績から生後7日目までにおけるBcl-2mRNA発現の増加は神経細胞系の発達に関わるものと思われるが水頭症群は神経細胞死の発現が示唆された。「結語」水頭症発達期には大脳皮質でアポトーシスの発現が示唆された。尚,本研究成績の一部は現在,国際誌に投稿中である.
