Effects of the intestinal tract on carbohydrate metabolism was stueied in respect of its direct action on the metabolism in the liver and of the indirect effect through the pancreatic endocrine functions.1. direct effects(1) In the jejunectomized rats which had been nurished for a week by total parenteral nutrition the response of plasma glucose and IRI to an intraperitoneal glucose load was significantly higher than in the controls.(2) Effects of various gut hormones on cAMP production, phosphorylase activity and glucose output were investigated in primary cultured hepatocytes. Glucagon, secretin and VIP augmented these three parameters, but others such as GRF, GIP and PHI did not stimulate all three.2. A new candidate for incretin-like substance and a physiological significance of the enteroinsular axis. In response to an oral glucose load the plasma GLP-1 immunoreactivity level was increased in man and GLP-1 stimulated IRI release from the perfused rat pancreas. This might be the new incretin. It became clear that the enteroinsular axis acted on the pancreatic A cells as well on the basis of the fact that an increase in plasma glucagon level in response to arginin was augmented after an oral glucose load.
