鶏の主要組織適合複合体であるB座位を構成するF,L,Gの3領域の遺伝子について、その構造をDNAレベルで分析し、免疫能力並びに抗病性との関連性について検討した。試料のDNAは赤血球から抽出し、制限酵素で切断したのち、今回購入したミニチラーで冷却しながら電気泳動し、サザンハイブリダイゼーションにより、RFLP分析を行なった。以下に結果をまとめると:1.G領域に対するRFLPの標準パターンの作成。(1)制限酵素として、EcoRI,PvuII,HindIIIを使用するこにより血清学的にB座位がホモである12系統から得た8種のハプロタイプを織別することが出来た。(2)上記の他にB座位がヘテロである7種のハプロタイプを混合してRFLP分析するこにより、合計21本のバンドが検出され、各バンドの大きさを推定することにより、血清学的に同定されたハプロタイプに対する標準パターンを作成した。2.G領域が同一でF領域のRFLPパターンの異なる個体を用いた免疫能力並びに抗病性との関連性。(1)G領域がB^<21>ホモでF領域のRFLPパターンの異なる個体をもちいた場合、抗体産生能及び植片対宿主反応能とF領域のRFLPパターンの関連性については例数が少なく一定の傾向が見られなかった。今後は、例数を増やして検討する必要がある。(2)抗病性との関連性については、マレック病ウイルス接種試験により、発症した個体と末発症の個体のDNAをEcoRI,とHindIIIを用いて消化したRFLPパターンを比較した結果、特定のバンドの出現頻度に差が見られF領域との関連性が示唆されたが、今回の結果ではG領域が同一の例数が少なかったため、今回の結果でF領域が明らかになった系統を用いて更に検討する必要がある。

The main tissue fitness complex is composed of three domains: F,L,G, DNA analysis, immunity, and disease resistance. The DNA of the sample was extracted from red blood cells, cut off by restriction enzymes, cooled down, and analyzed by RFLP. The following results were obtained: 1. The preparation of RFLP standard in G domain. (1)The restriction enzymes EcoRI,PvuII,HindIII are used in the serological B locus. (2)In addition, 7 kinds of B loci were mixed and analyzed by RFLP. The results showed that 21 loci were detected, and the size of each locus was estimated. The standard B locus was determined by serology. 2. RFLP patterns in the same G domain and in the same F domain are different from each other. (1)G In the case of domain B, <21>the number of RFLP fragments in F domain is relatively small due to the correlation between antibody production energy and host reaction energy. In the future, the number of cases will increase.
