A Growth Inhibitor of Rat Hepatocytes Synthesized by a Rat Hepatoma Cell Line, FF101

大鼠肝癌细胞系，FF 101，建立在无血清，无蛋白质的培养基合成的生长抑制剂的成年大鼠肝细胞在原代培养。FF 101条件培养液的凝胶过滤显示出两个生长抑制活性峰，其分子大小分别为200 kD和10 kD以下，小分子生长抑制剂初步命名为FF-GI。FF-GI对大鼠肝细胞DNA合成有抑制作用，用胰蛋白酶、二硫苏糖醇处理和100 ℃加热30 min可使其抑制活性消失，FF-GI对酸处理和100 ℃加热5 min均稳定，提示FF-GI可能是一种新的肝细胞生长抑制剂。

A rat hepatoma cell line, FF101, established in serum-free, protein-free medium synthesizes a growth inhibitor of adult rat hepatocytes in primary culture. Gel filtration of FF101-conditioned medium disclosed two peaks of growth inhibitory activity with the molecular sizes over 200kD and smaller than 10kD.The growth inhibitor of smaller molecular size was tentatively designated as FF-GI.The DNA synthesis of rat hepatocytes was inhibited by FF-GI.The inhibitory activity of FF-GI was abolished by treatments with trypsin, dithiothreitol and heating at 100ﾟC for 30min, although it was stable against acid treatment and heating at 100ﾟC for 5min.These results indicate that FF-GI may be a novel growth inhibitor of adult rat hepatocytes.
