We studied on the effects of photoradiation therapy(PRT) using the Argon laser tuned to 514.5 nm in the rat C6 glioma model and clinical patients. Our results from the study of the rat C6 glioma model with hematoporphyrin derivative(HpD) indicate that the effects of PRT on tumors include both tumor cell damage and endothelial damages of tumor vessels, and these are the main mechanisms causing tumor destruction. We have obtained clinical results from 6 patients treated with HpD and one patient most recently treated with Photofrin II.In the former group, there was no marked effect of PRT on malignant gliomas, in terms of time interval to tumor relapse and survival period. However, for the latter patient, there seems to have been a remarkable effect of PRT, indicated by MRI and CT, and histopathological findings of the specimen obtained during the second surgery. Differences of treatmant method from the 6 patients were (1)the use of Photfrin II, (2)higher power of laser, and (3)macroscopically total removal of tumor. Clinical results suggest that a new photosensitive substance, Photofrin II, may be a potent drug for treatmant of malignant glioma by means of PRT.Further investigations should be necessary to confirm these results.
