アミノ酸としてグリシン(Gly、C_2H_5NO_2、分子量57)を、また医薬品としてインドメタシン(IMC、C_<19>H_<16>CINO_4、分子量358)を取り上げ、標題の検討を行った。1.Glyではa形(準安定)とγ形(安定、望まし)、また、IMCではa形(準安定)とγ形(安定、望まし)、β形(擬多形)について、溶解度およびみかけの溶解熱を求めるとともに、結晶の同定と含有比の測定をGlyではIR、IMCではDSC、XRDで行った。2.IMCのγ形からβ形への固相内転移に及ぼず溶媒(EDC)ならびに混合結晶中の初期β形含有比Γ_<β,0>の影響を5℃、Γ_<β,0>=0〜0.7溶媒含有率η_<EDC>=80〜50%の条件で検討した。(A)β形含有の増大とともにγ形からβ形への転移が速やかに起こる、(B)Γ_<β,0>=0.3のとき転移速度はη_<EDC>=60%で最大値をもつ(C)転移速度はAvrami-Eroffeevの2次元式に従う、ことがわかった。これらのことから、この固相内転移は一種の溶媒媒介転移で、両多形の結晶界面で自触反応的に進行することが考察された。3.(1)Gly/水とIMC/EtOH系での種々の条件の過飽和溶液からのa形とγ形の晶折挙動を検討し、次の結論を得た。(A)いずれも飽和度の高い領域では、準安定のa形が発生した。(B)両多形の溶解度の比,ζが小さいほど、温度が高いほど、両多形が選択的に析出する領域の境界の過飽和度,S_<α,b>が小さいことがわかった。(C)撹拌速度大、冷却速度大ほど、S_<α,b>が小さいことがわかった。(D)S_<α,b>を該化速度および、二次元該式長速度の理論式から展開して求めたが、実測値の傾向と一致した。(2)準安定と安定形の溶解度の間の濃度領域で、溶媒媒介転移を検討した。成長速度と溶解速度の速度式を独立して測定し、両式を合成することで溶媒媒介転移中の濃度の経時変化を求めた。実測値の傾向が得られることがわかった。
