It has been suggested that cells from the periodontal ligament have the potential to create new connective tissue attachment. The periodontal ligament cells populated limitedly on the root surface of the wound in vivo due to epithelium down growth. We have been developing a new periodontal regeneration technique for the teeth with reduced periodontal ligament using the combination of tissue culture and transplantation of the teeth.The purpose of this study was to examine effects of enamel matrix derivative (EMD) application on periodontal healing after implantation of teeth with cells populating from periodontal ligament in vitro. After the Incisors from beagles were extracted, the periodontal ligament and cementum at 5 mm from coronal part of the roots were removed and the root surfaces were planed. The periodontal ligament in the apical part of the root was retained. Following the etching by 24 % EDTA on the root planed surface, Emdogain was applied on the root planed surface. After the teeth were incubated in a-MEM with antibodies and 15% fetal bovine serum for 6 weeks, the teeth were transplanted. Eight weeks after transplantation, the specimens were prepared for histological analysis.Most of the root planed surfaces of Emdogain application group were observed new cementum with fibers. It was concluded the enamel matrix derivative could increase the area of ALP-positive cells and enhance cementum formation after transplantation of teeth cultured in vitro.
有人认为,牙周膜细胞有潜力创造新的结缔组织附着。牙周膜细胞由于上皮向下生长,在体内只能在创面根面有限地增殖。本研究采用组织培养与牙移植相结合的方法,研究牙釉质基质衍生物(elaminalmatrixderivatives,EMD)对牙周组织再生的影响。拔除比格犬切牙后,去除距牙根冠部5 mm处的牙周膜和牙骨质,刨平牙根面。保留牙根根尖部的牙周膜。在根平面上用24%EDTA进行蚀刻后,将Emdogain应用于根平面上。将牙齿在含有抗体和15%胎牛血清的a-MEM中孵育6周后,将牙齿移植。移植8周后取标本进行组织学分析,Emdogain组根面平整后大部分可见新生牙骨质,纤维化。结论:釉基质衍生物可以增加体外培养牙齿移植后碱性磷酸酶阳性细胞的面积,促进牙骨质的形成。
