The synthesis of the human sperm factor -recombinant PLC zeta- and its clinical application.

受精时，卵母细胞被“精子因子”激活。最近，精子特异性磷脂酶C（PLC），被称为PLCzeta，被报道为精子因子的候选者，因为PLCzeta cRNA和重组PLCzeta蛋白可以引起小鼠卵子中的钙振荡。在本研究中，我们利用真核表达系统合成了重组人PLCzeta蛋白（re-hPLC zeta）。将纯化的re-hPLC β-D微注射到小鼠卵中，并使用共聚焦激光显微镜记录细胞内钙离子水平的变化。结果，在卵中观察到钙振荡，表明re-hPLC具有促性腺激素活性。如果我们正在进行的研究证实了re-hPLC的安全性，则re-hPLC可用于激活失败病例的胞浆内单精子注射程序。

At fertilization, the oocytes are activated by "sperm factor". Recently, the sperm-specific phosphor lipase C (PLC), termed PLCzeta, is reported as a candidate of the sperm factor because the PLCzeta cRNA and recombinant PLCzeta protein can evoke the calcium oscillations in murine eggs. In the present study, we have synthesized the recombinant human PLCzeta protein (re-hPLCζ) using eukaryote-expression system. The purified re-hPLCζ was microinjected into murine eggs and the changes in the levels of the intracellular calcium ion were recorded using conforcal laser microscopy. As a results, the calcium oscillations was observed in the eggs, indicating that the re-hPLC had a oscillogen activity. If the safety of the re-hPLC is confirmed by our on-going study, the re-hPLC can be used for the intracytoplasmic sperm injection procedure of the cases of activation failure.
