A system consisting of an ultra-high-performance liquid chromatography system and an electrospray triple quadrupole/QTRAP mass spectrometer (UHPLC-ESI-TQ / QTRAP-MS/MS) shall be procured. The UHPLC system should include a binary high-pressure pump with a pressure limit of 1200 bar, a heated column oven and a thermostated autosampler. The TQ/QTRAP mass spectrometer should have high selectivity as well as high sensitivity for the detection of metabolites in MRM (Multiple Reaction Monitoring) mode to enable targeted quantitative analysis of low-abundant metabolites and to avoid interferences with matrix background. As additional options to optimize the separation of target analytes from matrix background and isobaric compounds, the TQ/QTRAP mass spectrometer should have an upstream ion mobility and the option to use a second generation of productions for quantification (MRM3). Furthermore, the possibility of tandem mass spectrometry (MS/MS) should be given within an MRM experiment to confirm the identity of target analytes. In addition, powerful software tools for processing and interpreting the mass spectrometry data are an important part of the overall system.The system applied for is intended to be used for the targeted quantitative analysis of small organic molecules in complex biological samples. One focus is the targeted quantitative analysis of phytohormones in plant model organisms and cultivated plants. On one hand, the results should lead to a better understanding of the signalling pathways that are involved in the adaptation of these plants to different biotic interactions and, on the other hand, it should be elucidated how plants integrate different environmental signals that are associated with stress. Furthermore, the role of phytohormones and other substances (saponins, lipids and flavonoids) in cultivated plants in relation to abiotic stress factors such as salt content in the soil, water deficiency, heat or cold will be investigated. The aim is to gain knowledge of plant signalling pathways and adaptation mechanisms, which should lead to more efficient cultivation of crops.Furthermore, the system applied for is intended to be used for is the quantitative analysis of primary and secondary metabolites in the blood, milk and the intestinal contents of farm animals (cows, chickens, pigs). The aim of these investigations is to characterize the effects of feeding, husbandry and management on the metabolism and the microbiome of livestock and thereby identify biomarkers for the metabolic health of livestock. These results should form the basis for resource-saving and animal welfare food production.
