Sensing of intracellular analytes (mostly located in endosomes/lysosomes) will be performed by loading cells with analyte-responsive dyes, optionally coupled to carrier particles, via endocytosis, microinjection, or membrane permeation. Intracellular analyte concentrations will then be determined by multiplexed read-out of the dye signals. In order to avoid crosstalk with the potential (here unwanted) pH-response of the dyes, the analyte-response of the dyes will be read-out laterally resolved either in the form of time-resolved fluorescence microscopy (FLIM) or surface-enhanced Raman scattering (SERS) microscopy. Multiplexed read-out of dyes in different cells within two- and three-dimensional co-culture systems will be facilitated by cell-labelling with SERS tags.
