The defect of amino acids in the lens may affect the metabolism and cause to alterations of the structure and functions of lenses. The purpose of this project is investigate the effect of amino acid deficiency on function of the lens. 1. Gamma-glutamyl cycle play a important role on amino acid transport to lens. 2. The amino acid transport to lens wa decreased with aging, but free amino acid concentration was increased. By adding inhibitors of <gamm>-glutamyl cycle enzymes to cultured lensees, free amino acid concentration was also increased. 3. The activity of lens LDH was not affected by inhibitors, yet <gamm>-GTP was inhibited. 4. Effect of inhibitors of amino acid transport was not found out in in vivo experiments. But it was suspected that effect of aqueous humor collection lead lens cloudy. It was suggested that lowering of amino acid transport in lens epithels was related of lens function by lowering amino acids uptake and generation of cataract should be dissolved. From the result of in vivo experiment, chemical and physical changes of aqueous humor (e.g. content and change of tension) also should be considered.
晶状体中氨基酸的缺陷会影响晶状体的代谢,引起晶状体结构和功能的改变。本研究旨在探讨氨基酸缺乏对晶状体功能的影响。1. γ -谷氨酰循环在氨基酸转运到晶状体中起重要作用。2. 随着衰老,晶状体氨基酸转运量减少,游离氨基酸浓度增加。在培养的晶状体中加入< γ - >-谷氨酰循环酶抑制剂,游离氨基酸浓度也有所提高。3. 抑制剂对晶状体LDH活性无影响,但对< γ - >-GTP有抑制作用。4. 体内实验未发现氨基酸转运抑制剂的作用。但怀疑是房水积液导致晶状体浑浊的影响。提示晶状体上皮内氨基酸转运的降低与晶状体功能有关,可能与晶状体对氨基酸的摄取减少和白内障的发生有关。根据体内实验的结果,还应考虑房水的化学和物理变化(如含量和张力的变化)。
