项目摘要

ヒト皮内より発生した悪性線維性組織球腫(原発巣)の組織片から、組織培養(イーグルMEM液、仔牛血清10%)において細胞株を樹立した。現在(1989年1月)2.5年間に64代の継代培養を続けている。もとのヒトでの病理組織学的細胞形態は類上皮細胞を主体とし多核巨細胞を混ぜ、紡錘形細胞は少数であった。組織学的に腫瘍塊内の血管増生が目立った。組織培養された細胞は、その形態が非常に多彩であった。すなわち、長紡錘形(SS)、紡錘形(PS)、類上皮形(Epi)、多核巨細胞(MNGC)、リンパ球様小球形細胞などであった。これらの細胞からその形態に応じ細胞のクローニングを行った。すると興味あることにSS細胞は集団をなして培養上で車輪状構造をつくり、PS細胞は花ムシロ状構造をつくった。これらはヒトの隆起性皮膚線維肉腫の病理組織像でみられるものと同一であった。また類上皮細胞は個々の細胞では紡錘形を示したが、細胞同志が接触することによりこれらの細胞が類上皮形に変化した。そして数多くの多核巨細胞がこれらの類上皮細胞群の中に見出された。以上のことは隆起性皮膚線維肉腫は悪性線維性組織球腫の一型(紡錘形細胞型)であることを示している。また多核巨細胞の多くは類上皮形細胞に由来することが判明した。今後、ヌードマウス内(in vivo)の系において、これらの細胞がいかなる性質をもっているか研究されることが望まれる(1989年度、科学研究費申請中)。

The origin of multinucleated giant cells and epithelial-like cells has been clarified.
