1.目的 本研究は、感情障害家系のRFLPによる遺伝子連鎖分析を行い、感情障害の責任遺伝子座位をあきらかにするとともに、Egelandらの結果ないしGurlingら、Gershonらの結果の検討を行うことを目的として立案された。2.対象と方法 対象・以下の手続によって選び出された発端者とその家族を対象とした。(1)昭和63年4月1日より5月31日の間に帝京大学医学部附属病院精神科外来を受診した患者の内からカルテ記載によって感情障害と診断され、かつ第三度親族内に2名以上の精神障害者を持つ患者を選び出す。(2)これらの患者について、各担当医がRDCによる診断を行ない、感情障害と診断された患者を選び出す。(3)これらの患者とその家族に対して、研究の趣旨を説明し、研究の同意を得る。同意を得られた者を発端者とする。以上の手続によって選ばれた三家系16名(罹患者7名、非罹患者16名)を対象とした。方法・これら16名について説明、採血を行い白血球DNAを抽出し制限酵素PvuII、SacIで切断、通常のサザンブロット法によって11番染色体短腕末端部にあるインスリン遺伝子及び癌遺伝子Ha-ras-1のプローブを用いてRFLPを分析した。現在これらの結果を用いて罹患者との連鎖分析、検定を行っている。しかし、統計的検定を行うにたる症例数には至らなかった。なお、来年度以降さらに症例を蓄積し、また11番染色体以外のCandidate geneについても分析を行う予定である。

The restriction enzyme PvuII was cut off by SacI. The terminal part of the short wrist of 11 polyploid chromosomes was analyzed by RFLP analysis. At present, the results of this study are based on the analysis of the relationship between patients and patients. According to the statistics and statistics, the number of cases was significantly different from one to the other. In the coming year, in order to reduce the number of cases of illness, the number of cases in which the symptoms were stored and 11 times more than the chromosomes of the Candidate gene were analyzed.
