正常移行上皮へのBROMODEOXYURIDINE(BRDU)の膀胱内注入による取り込みを見るため、まず雄の成犬9頭にBRDUを投与した。投与方法は最初の7頭にはBRDU750mgを生理食塩水50mlに溶解し膀胱内に注入し、のこりの2頭にはその1/2の濃度で投与したまこの際濃度が変化しないように両側の尿管は結紮した。投与約2時間後に膀胱を摘出し膀胱の後壁、両側壁より標本を採取し、70%アルコールにて固定後、パラフィン包埋し抗BRDUモノクロナール抗体を用いABC法にて染色した。移行上皮細胞1000個中の標識細胞数を数えLABELING INDEX(L、I)で表した。標識細胞は重荷中間層に認められ、L.I.は平均9、7で濃度による差は見られなかった。次に膀胱移行上皮癌のため膀胱全摘術を予定された患者に対し、手術直前にBRDUを投与した。投与量はBRDU1500mgに生理食塩水50mlとした。投与時間は平均5時間で、膀胱摘出後腫瘍及び正常部を採取し前期の方法と同様に染色しL.I.を算出した。患者は3名で術後の病理組織診断は微小浸潤を伴う上皮内癌1例であった。正常のL.I.は平均7、8で、標識細胞は高座層から中間層にかけて認められた。上皮内癌の症例においてはL.I.は24.7で標識細胞はほぼ全層に平均してみらた。上皮異形成例、移行上皮癌例は腫瘍が小さく判定が困難であった。

Normal transitional epithelium of BROMODEOXYURIDINE(BRDU) was injected into the bladder. Method of administration: BRDU750mg was dissolved in 50ml of physiological water and injected into the urinary bladder. The concentration of BRDU 750 mg was 1/2 of that of BRDU 750 mg. About 2 hours after the injection, the bladder was extracted, and the posterior wall and lateral wall of the bladder were taken. After 70% of the bladder was fixed, the anti-BRDU antibody was embedded and stained by ABC method. Number of transitional epithelial cells labeled out of 1000 LABELING INDEX(L, I) The marker cells are heavily charged with intermediate layers, and L.I. concentrations average 9 and 7. For transitional cell carcinoma of the urinary bladder, complete cystectomy was performed to determine the patient's position and the BRDU was administered before surgery. The dosage is BRDU1500mg and physiological water 50ml. The average time of injection is 5 minutes. The bladder is removed and the normal part is taken. The method of staining is the same. Three patients were diagnosed as microinvasive intraepithelial carcinoma by postoperative pathology. Normal L.I. average 7, 8, marked cells In the case of intraepithelial carcinoma, the L.I. is 24.7 and the marker cells are 24.7. Epitheliosis and transitional carcinoma are difficult to determine.
