项目摘要

Amplification and expression of oncogenes were investigated in 55 cases of human ovarian cancer tissues. Gene amplificaion of unknown nature was screened by in-gel renaturation assay and amplification of the c-myc, Ki-ras and neu oncogenes were determined by Southern blot hybridization. No significant gene amplification was found in all the cases. By Northern hybridization assay, the c-myc gene was overexpressed in 29 cases (52.7%) whereas either k-ras or neu was not.The overexpression more than 3-fold of the c-myc gene was significant in serous cystadenocarcinoma and endometrioid carcinoma tissues compared with other histological types of tumor. Immunohistochemistry and in-situ hybridization revealed that tumor cells overexpressed both the c-myc gene and the protein.Although the expression level was not related to grading of malignancy, it is suggested that the c-myc gene is involved in early stages of carcinogesis of ovarian tumors.
