We studied the changes of anionic sites produced by preservations in basement membrane of lungs using Wister rats.They were divided into 4 groups as follows ;A group : controlB group : stored at room temperature in heparinized salineC group : stored at 37゚C in heparinized salineD group : stored at 37゚C in EP3 solutionSmall tissue blocks were taken from the lungs and anionic sites were stained using strong cationic polyethyleneimine as a tracer following Schurer's method. After staining of anionic sites, we observed the sections on. electrone microscopy and caliculated the dot density of basement membrenes(dot number / cm at the magnification of 15410)and analyzed them by-the system(Hard : NEC 9801 VX, Soft : Nikon Cosmozone).RESULTSA group : 10.2<plus-minus>1B group : 12.6<plus-minus>1C group : 12.0<plus-minus>1D group : 11.8<plus-minus>1.8 (mean<plus-minus>S.CONCLUSIONCompared to the A group(control), B, C and D group hdd higher density of anionic sit significantly(p<0.05)We need more experiments to put an exact physiological interpretation on these results.
本实验采用Wister大鼠,观察了肺基底膜内阴离子位点的变化,实验分4组,A组:对照组,B组:室温肝素化生理盐水保存组,C组:37 ℃肝素化生理盐水保存组,D组:从肺中取出小的组织块,用强阳离子聚乙烯亚胺作为示踪剂对阴离子位点进行染色,Schurer的方法。阴离子位点染色后,在电镜下观察,计算基底膜的点密度(15410倍下点数/ cm),并用NEC 9801 VX系统(硬)和NikonCosmozone系统(软)进行分析,结果表明:与A组(对照组)相比,B组:<plus-minus>10.21B组:<plus-minus>12.61C组:<plus-minus>12.01D组:<plus-minus>11.81.8(均数<plus-minus>S)。C、D组阴离子位点密度显著高于对照组(P<0.05)。
