近年の遺伝子工学の急速な進展に伴い,リンパ球やマクロファ-ジなど種々の免疫担当細胞が産生し,細胞間の情報伝達を行うインタ-ロイキンの働きがしだいに明らかになってきた.そこで本研究では,根尖性歯周炎罹患患者から末梢血や病変部組織を採取してリンパ球を抽出し,その活性の異常の有無をリンパ球サブセットとILー2活性,ILー2R発現との関連を調べることにより明らかにした.また根管内浸出液中のILー2活性はELIZA法で測定した.さらに根管内からの分離頻度が高いB.melaninogenicusに対する宿主の免疫応答を,本菌の刺下で培養したリンパ球サブセットとILー2活性およびILー2R保有細胞の変動と関連づけながら調べた.1.被検者:根尖性歯周炎罹患患者[全身状態が良好で,単根歯の根尖部にX線透過像を有し外科的処置の対象となるもの(対照群),および通常の処置法で治療が可能なもの(実験群)…各10名],2.試料の採取:臨床診査,X線診を行った後,各グル-プの被検者から末梢血,根管内浸出液,病変部組織(対象者のみ)を採取した.3.リンパ球の抽出,解析およびILー2活性の測定:2.で得た試料のうち病変部組織ではコラ-ゲナ-ゼ処理した後,密度勾配遠心法でリンパ球を抽出し,抗T細胞,抗B細胞,抗ILー2R,CD4,CD8の各モノクロ-ナル抗体で染色し,フロ-サイトメトリ-で解析した.また根管内浸出液のILー2活性はELIZA法で測定した.その結果,対照群に比べ実験群では,とくに病変部組織においてリンパ球機能の異常を示唆する結果が得られたが,末梢血と根管内浸出液に関しては,現時点では有意の差を見出せなかった.さらに症例を増やし検討を進めている.

In recent years, rapid progress has been made in genetic engineering, and the production of immune cells and the transmission of information between cells have been carried out. In this study, the relationship between IL-2 activity and IL-2R expression was investigated in patients with apical periodontitis. IL-2 activity in root canal extract was determined by ELIZA method. B. melaninogenicus is the host's immune response, and the culture of this bacterium under the root canal maintains the cell's dynamic association with IL-2 activity. 1. The host: Patients suffering from apical periodontitis [who are in good general condition, have X-ray transmission images of the apical part of the root canal and are suitable for surgical treatment (control group), and are suitable for treatment with normal treatment methods (actual group)... 10 each], 2. Sample collection: After clinical examination, X-ray examination, each group of patients will be examined for peripheral blood, root canal extract, diseased tissue. 3. Extraction, analysis and determination of IL-2 activity: 2. After treatment, density matching and remote-control method were used to extract the samples. Anti-T cells, anti-B cells, anti-IL-2R, CD4, CD8 antibodies were stained and analyzed. IL-2 activity in root canal extract was measured by ELIZA method. The results showed that the root canal extract was significantly different from that of the peripheral blood. Today's symptoms are increasing and discussions are progressing.
