The deposition of amyloid beta peptide (Ab) is among the defining features of Alzheimer's Disease (AD). Accumulation of Ab may occur as a consequence of enhanced production or altered catabolism of Ab. recently, significant progress has been achieved regarding the determination of the mechanisms leading to the generation of Ab. In contrast, virtually no information has been available as to whether and how Ab is cleared by cells. We have shown that Ab peptides, 40 to 43 amino acids in length, are ligands of the LDL receptorrelated protein (LRP) and heparan sulfate proteoglycan (HSPG) pathway, which is also involved in the clearance of apolipoprotein (apo) E, a major cholesterol transporting protein in the central nervous system. Ab and apo E compete with each other for the binding to LRP when present in a soluble form. Prolonged exposure of Ab to apo E containing lipoproteins leads to the formation of Ab/apo E complexes. These complexes are avidly internalised by LRP and HSPG, but are completely resistant to lysosomal degradation. It will be the specific aims of the proposed project a) to identify the domains of LRP responsible for Ab binding, b) to elucidate the roles of other LDL receptor-like molecules in the uptake of Ab, and c) to follow the subcellular trafficking of Ab internalised by these cell surface molecules. Zusammenfassung: Die Ablagerung des Amyloid b Peptids ist eine der morphologischen Korrelate der Alzheimer Demenz. Die Akkumulation von Ab scheint die Folge einer erhöhten Produktion oder eines veränderten Katabolismus von Ab zu sein. In jüngster Zeit wurden neue Erkenntnisse zur Aufklärung des Mechanismus der Bildung von Ab gewonnen. Im Gegensatz dazu gibt es kaum Daten zur Frage, ob und wie Ab von Zellen abgebaut wird. Wir konnten zeigen, daß Ab Peptide (mit Länge von 40 bis 43 Aminosäuren) Liganden des LDL receptor-related protein (LRP)/Heparan sulfate proteoglycan (HSPG) pathway sind, der auch beteiligt ist am Abbau von Apolipoprotein (Apo) E. Apo E ist hauptverantwortlich für den Cholesterintransort im zentralen Nervensystem. Ab und apo E konkurrieren um die Bindung an LRP, wenn beide in gelöster Form vorliegen. Wird Ab über längere Zeit mit Apo E enthaltenden Lipoproteinen inkubiert, führt dies zur Bildung von Ab/apoE-Komplexen. Diese Komplexe haben eine hohe Affinität zu LRP und HSPG, werden internalisiert, aber nicht lysosomal degradiert. Ziele des beantragten Projekts sind: a) die für die Ab Bindung verantwortlichen Domäne des LRP zu identifizieren, b) aufzuklären ob andere LDL-Rezeptor-ähnliche Moleküle an der zellulären Aufnahme von Ab beteiligt sind, und c) den weiteren intrazellulären Weg des aufgenommenen Ab aufzuklären.
