研究実施計画にもとずき本年度は以下の研究を行った。(1)mdx-nude mouseの作製本研究に最適なホストであるmdx-nude mouseは ジストロフィン欠損mdx mouseとnude mouseを交配することにより、両形質を共有するmouseとして作製することができる。すでに委託生産を開始し、本年度も実験に必要なmouse飼育し必要に応じて供給を受けた。研究費の大半がこれにあてられた。(2)種々設定条件下での正常骨格筋のmdx-nude mouseへの移植基本的にmdx-nude mouseと同系の,C57BL control mouse下肢正常骨格筋を無菌的操作のもとに細切、トリプシン、コラゲナーゼで処理しホモジェネートとしてmdx-nude mouse下肢伸筋内に注入した。この際、遺伝子の移入と融合を促進させるために、上記基本的操作に加えて、塩素ブピバカインやコラゲナーゼによるホスト筋の前処置などを計画したが本年度は結果的に上記基本操作までの実験にとどまった。(3)ジストロフィン蛋白および遺伝子の発現状態の検討免疫組織化学とin situ hybridization(ISH)によりジストロフィン蛋白とそのmRNAの発現状態を検討した。現在のところ実験の技術的問題の解決に期間を要しており一定の結論を得ていない。
