项目摘要

回転式全胎仔培養装置で,胎齢12.5日目のSprague-Dawley系ラット胎仔を同系ラットから採取した迅速遠心血清に人工培養液を混じたものを培養液とし,5%二酸化炭素,95%酸素放流下に24時間培養を行った.培養開始6時間後に心拍動と血液循環のみられる胎仔を暴露対象とし,コントロール(38℃)群,ニコチン1560μM添加(38℃)群,コチニン1560μM添加(38℃)群,メタンフェタミン5μM添加(38℃)群,および42℃群の5群を作成し18時間追加培養を行った.胎仔培養終了後,各群胎仔につき形態学的異常の有無を検討するとともに,生存例につき頭殿長[mm],頭部長[mm],体節数[個]を測定のうえ,各群の抑制形態の違いを頭部長/頭殿長比,頭殿長/体節数比を用いて比較検討した.明らかな形態学的異常は認められず,各群の頭部長/頭殿長比はそれぞれ,0.60±0.02,0.59±0.03,0.06±0.03,0.60±0.02,0.59±0.02,頭殿長/体節数比はそれぞれ,0.17±0.01,0.17±0.02,0.17±0.01,0.17±0.02,0.16±0.02と有意差は認められなかった.以上から,42℃高体温環境下,ニコチン,コチニン,メタンフェタミン添加のいずれにおいても,対照群に比べ発育抑制をうけるのであるが,これら,暴露の種類いかんにかかわらず,この時期においては外胚葉系のバランスは保たれてsymmetricalに発育が抑制されるものであることがわかった.
The whole fetus culture system was established in a reverse-flow whole-fetus culture apparatus. The embryos were cultured for 12.5 days in Sprague-Dawley system. The embryos were cultured for 24 hours in a 5% diacidified carbon and 95% acid release medium. 6 hours after the start of culture, the fetus was exposed to heart beat and blood circulation, and 5 groups were prepared for additional culture for 18 hours. At the end of fetal culture, morphological abnormalities in each group were examined. In survival cases, head length [mm], head length [mm], and number of body segments were measured. In each group, head length/head length ratio and head length/number of body segments ratio were compared. The ratio of head length to temple length was 0.60 ± 0.02, 0.59 ± 0.03, 0.06 ± 0.03, 0.60 ± 0.02, 0.59 ±0.02, and the ratio of temple length to body segment number was 0.17 ± 0.01, 0.17 ± 0.02, 0.17 ± 0.01, 0.17 ±0.02. 0.16±0.02 intentional difference. Under the high temperature environment of 42 ℃, the temperature of the plant is higher than that of the control group, and the development of the plant is inhibited.
