本年度は、本研究で扱う特定の哺乳類の遺伝子を、試料中に含まれる細菌やカビなどの遺伝子の中から特異的に効率よく増幅・単離するための系を作った。目的の生物由来の遺伝子配列を調べるための領域として、ミトコンドリアDNAのチトクロムb遺伝子を選定した。古いDNAは、重度の損傷を受けて断片化しているために、PCR手法により増幅できる遺伝子断片の長さは一般的に短いものに限られるが、この領域ならば短い領域でも生物種についての情報を得やすい。哺乳類ならばほとんどの種類で共通である配列のほか、ウサギ、シカなどの生物種ごとに特異性が大きい配列をいくつか選定してプライマーセットを作製した。現世の哺乳類の毛や毛皮、肉などからDNAを抽出して遺伝子増幅を行い、これらのプライマーセットの有効性を検討した。その結果、哺乳類共通のプライマーのほかに特定の種に特異的なPCRのプライマーセットをいくつか得ることができた。これらのプライマーを組み合わせて検討すれば、ヒトの遺伝子などのコンタミネーションをほぼ確実に区別できる。現在、古代試料として古墳から出土した青銅器に貼付されていた毛皮からDNAを抽出し、毛皮の生物種を同定すべく実験を進めている。本試料は、青銅器に加工された形で出土したため、多量のさびがついており、金属や金属イオンをいかに除去して目的のDNAを精製するかが大きな課題であった。しかし、このほど生物種特異的なPCR増幅産物が得られたので、現在そのDNA領域を調べている。
