本研究は、1992年 Allenらによって明らかにされたアンドロゲン受容体遺伝子塩基配列をもとに、女性の約90%に保存されているCAG繰り返し配列部分をPCR法によって検出することにより、皮膚腫瘍において良性、境界型、悪性に分類されているものについてそのモノクローナリテイの有無を検出することにより、腫瘍細胞の単一性という面から再評価することを目的とする。本年度はまず、安定した実験方法の確率をめざした.8例の女性メラノーマ患者より得られたホルマリン固定パラフィン包埋組織より腫瘍部分と非腫瘍部分を切り出しDNAを抽出後、サンプルの一部を制限酵素Hhalで処理した。次にアンドロゲン受容体遺伝子のCGA繰り返し配列部分をはさむ形で作成したプライマーを用いて、制限酵素処理後のサンプルと未処理のサンプルについてPCR法で増幅を試みた。 しかし、当初期待していた程度のDNAの増幅が得られなかった。 メラニンによる阻害作用あるいはサンプル内に混入しているポリメラーゼ阻害物質の存在が疑われたためキレートビーズなどによるサンプルの洗浄を行いインターナルコントロールの増幅は可能となったが、アンドロゲン受容体遺伝子の増幅が不良(8例中1例のみ増幅した)のため、さらにプライマー位置を変更し再度増幅を試みている。また増幅された1例のサンプルについてはプライマーの一方のみを標識しPCR法による増幅後ポリアクリルアミドゲル上での泳動を行っている。
