第1に、これまで行ってきた、ヒロ卵における受精時細胞内カルシウム増加反応(受精カルシウム反応)の解析を進めた。方法としては、体外受精・胚移植にて未受精に終わった卵と夫精子とを、患者夫婦の同意を得た後に材料として用い、卵の透明帯を酵素処理にて除去した後に、カルシウム結合制御蛍光色素であるFura 2をAM体の形で卵内に取り込ませ、特定波長をもった異なる2種類の紫外線を照射し、励起される蛍光の強度の比の変化から細胞内カルシウム濃度の変化を観察した。この方法により受精カルシウム反応の出現と、事前に行った体外受精・胚移植における受精率との間に相関が認められるか否かを検討した。体外受精・胚移植における受精率を、未受精群(A群)・受精率の低い群(B群)・受精率の高い群(C群)の3群に分類し、それぞれの群における受精カルシウム反応の出現率をみると、A群では出現率は低く、逆にC群では高く、B群では両者の中間の値をとった。すなわち、受精カルシウム反応の出現率と体外受精・胚移植における受精率とには相関が認められた。体外受精・胚移植における受精率は、精子受精能により規定されるところが大きく、従って受精カルシウム反応は精子受精能の新しい評価法となると考えられた。第2に、別な精子受精能の評価法として、精子のアクリジン・オレンジ染色法の検討を行った。その結果、本法による染色パターンと、体外受精・胚移植における受精率との間に相関が認められた。従って本法も精子受精能評価法として有用であると判断された。以上2つの方法を採用することにより、精子受精能評価がより正確に行えるようになうと期待される。
