The prognosic value of the transcription of tumor growth-related genes in endoscopic biopsy specimens from patients with malignancies of digestive tracts

肿瘤活检标本是在胃癌术前内窥镜检查中获得的。提取每份活检标本的总RNA，用逆转录聚合酶链式反应(RT-PCR)分析肿瘤生长相关基因的转录。细胞周期相关因子为细胞周期相关因子，尿激酶型纤溶酶原激活物和72-kd IV型胶原酶为侵袭相关因子，血管内皮生长因子和血小板衍生生长因子-A为血管生成相关因子，转化生长因子b和白介素10为免疫抑制相关因子。(2)PDGF-A和TGF-b是术前判断预后的指标。(3)术前评估每种癌的恶性潜能可能有助于对胃癌进行更合理的分类。

The tumor biopsy specimens were obtained during preoperative endoscopy of patients with gastric carcinoma. The total RNA from each biopsy specimen was isolated and a reverse trascriptase-polymerase chain reaction analysis (RT-PCR) was used to analyze the transcription of tumor growth-related genes. The factors examined included cyclin D1 and cyclin E as cell cycle-related factors, urokinase-type plasminogen activator and 72-kd type IV collagenase as invasion-related ones, vascular endothelial growth factor and platelet-derived growth factor-A as angiogenesis-related ones, and transforming growth factor b and interleukin-10 as immunosuppression-related ones.Results :(1) The incidence of PDGF-A and TGF-b mRNA expression was preoperative indicator of lymph node involvement in early and advanced gastric cartcinomas, respctively.(2) PDGF-A and TGF-b were preoperative prognostic indicators.(3) Preoperative evaluation of malignant potential of each carcinoma may lead to a more reasonable classification for gastric carcinomas.
