项目摘要

The tumor biopsy specimens were obtained during preoperative endoscopy of patients with gastric carcinoma. The total RNA from each biopsy specimen was isolated and a reverse trascriptase-polymerase chain reaction analysis (RT-PCR) was used to analyze the transcription of tumor growth-related genes. The factors examined included cyclin D1 and cyclin E as cell cycle-related factors, urokinase-type plasminogen activator and 72-kd type IV collagenase as invasion-related ones, vascular endothelial growth factor and platelet-derived growth factor-A as angiogenesis-related ones, and transforming growth factor b and interleukin-10 as immunosuppression-related ones.Results :(1) The incidence of PDGF-A and TGF-b mRNA expression was preoperative indicator of lymph node involvement in early and advanced gastric cartcinomas, respctively.(2) PDGF-A and TGF-b were preoperative prognostic indicators.(3) Preoperative evaluation of malignant potential of each carcinoma may lead to a more reasonable classification for gastric carcinomas.
