There is no reported vascular prosthesis that has the ability to biologically fit the host, grow and remodel. Lack of such grafts limits the long-term benefits for patients especially pediatric ones.As a first step toward the ideal graft, we examined a novel vascular prosthesis that consists of acellular matrix to evaluate the degree of the host tissue regeneration on our prosthesis.A porcine carotid artery with a diameter of 6-8mm was treated with 1 % detergent Triton X-100 for 48hours. The graft surface was heparinized. This acellular vascular prosthesis interposed the abdominal aorta in 4 adult beagle dogs. Animals underwent angiography and were sacrificed 20 days and 3-51 weeks after the surgery, respectively.Microscopic and immunohistochemical examination revealed that the nuclei of the artery were completely removed from the prosthesis before implantation. All the implanted grafts were patent without the irregularity of its wall. At 3 weeks postoperatively, the internal lumen of the graft was covered with the endothelial cells except the mid portion. At 18 weeks, the entire surface of all the grafts was covered with the endothelial cells. The SEM examination revealed that the endothelial cells were lined on the luminal surface in the longitudinal direction of the graft.Regeneration of the host tissue may be accomplished using the novel acellular xeno-vascular prosthesis.
