I examined the responsiveness of acute myeloblastic leukemia (AML) cells for G-CSF by culturing the leukemia cells from 15 patients with or without G-CSF. I also examined the phosphorylation of intracellular proteins such as MAP kinase, JAKs, and STATs in the leukemia cells before or after the stimulation with G-CSF. I found that in the cells of which proliferation was stimulated by G-CSF, the above proteins were phosphorylated by the stimulation with G-CSF. Namely, we can infer the responsiveness for G-CSF by examining the phosphorylation much faster than in vitro culture assay. However, we could not infer whether G-CSF stimulated the self-renewal or terminal differentiation of the cells, according to the kind of phosphorylated proteins or intensity of the phosphorylation.
