The aim of this study is to accelerate the angiogenesis in necrotic bone by combining vascular bundle implantation and FGF-2 administrationTwenty-four Japanese white rabbits were used. A portion of rabbit iliac crest bone was removed as a free bone graft and frozen in liquid nitrogen to ensure complete cellular necrosis. A narrow hole was created in the bone and the graft was placed in the proximal thigh. In group 1, FGF-2 was injected into the hole at a single dose of 100 micrograms, and the saphenous artery and its venae comitantes were passed through the hole of the bone. In group 2, injection of saline into the hole and vascular bundle implantation was used as a control. Neovascularization around the vessel was evaluated at weeks 1 and 2 after vestmentNeovascularization was observed along the implanted vascular bundle. The vessel density gradually increased from week 1 to week 2 in both groups. At 1 week after surgery, the vessel density in group 1 (0.148) was significantly higher than in group 2 (0.050). At 2 weeks after surgery, the mean of the vessel density in group 1 was 0.375 and that in group 2 was 0. 194. This difference was also statistically significant. The length of vessels at 1 week after surgery in group 1 (0.862mm) was significantly higher than in group 2 (0.330mm). At 2 weeks after surgery, the mean of the vessel length in group 1 was 2.625mm and that in group 2 was 1.383mm. This difference was also statistically significantA local single injection of FGF-2 improved surgical angiogenesis in necrotic bone in this study.
