【目的】動物実験等によりダイオキシンが女性ホルモンであるエストロゲンの代謝を変化させるという報告があり、ダイオキシンの毒性との関連が指摘されている。そこで、自治体焼却施設労働者における血清中ダイオキシン類濃度と尿中エストロゲン代謝物濃度を測定し、その関連性を検討した。【方法】対象者は焼却施設に勤務する男性57名を選択した。質問紙調査により対象者の属性を調査した。血清中ダイオキシン類濃度を曝露指標とした。採取した血液を血清分離、脂肪抽出、脂肪分解、溶媒抽出、カラムクリーンアップを行い測定試料とし、高分解能ガスクロマトグラフ質量分析計を用いて測定した。尿中エストロゲン代謝物(エストロン(E1)、エストラジオール17β(E2)、2位水酸化エストロゲン…4種類、4位水酸化エストロゲン…3種類および16位水酸化エストロゲン…2種類;計11種類)を影響指標とした。採取した尿を固層抽出、脱抱合体反応、溶媒抽出、誘導体化反応、溶媒抽出、カラムクリーンアップを行い測定試料とし、高分解能ガスクロマトグラフ質量分析計を用いて測定した。【結果】対象者の血清中ダイオキシン、ジベンゾフランおよびコプラナPCB濃度は毒性等量で、それぞれ、12.9、12.4および13.6pgTEQ/g脂肪であった。単相関分析より血晴中ダイオキシン類濃度(TEQ値)と16位水酸化エストロゲンの一つである尿中エストリオール(E3)濃度との間に有意な正の相関がみられた。年齢、Body mass index (BMI)、喫煙本数、アルコール摂取量の影響を調節した共分散分析においても、ダイオキシン類濃度とE3との間に有意な正の相関がみられた。また、BMI、喫煙量、アルコール摂取量とエストロゲン代謝物との間にも相関関係がみられた。【まとめ】ヒトにおいてもダイオキシン類がエストロゲンの代謝に影響を与える可能性が示唆された。同時にエストロゲンの代謝にはBMI、喫煙量、飲酒の影響を受けることが示唆された。
