平成18年度は,実験群にimmobilization stressを与え,経時的に採血を行い,血中コルチコステロンを測定する拘束ストレス実験を行った。1.実験群母子分離群4群(1)早期分離12時間群(12E:6匹),(2)早期分離3時間群(3E:6匹),(3)後期分離12時間群(12L:6匹),(4)後期分離3時間群(3L:6匹))と(5)対照群(6匹)の計5群とした。2.拘束ストレスimmobilization stressラットの身体をテープでボード上に固定して実施した。拘束時間は30分間とした。3.採血とコルチコステロン測定採血は拘束ストレス直前とストレス開始15分後,30分後,60分後,120分後,180分後,240分後,300分後,360分後の計9回行い,遠心分離した血漿は冷凍保存し,蛋白競合法にてコルチコステロンを測定した。【結果および考察】早期分離群(12E群と3E群)では,拘束ストレス開始直後から血中コルチコステロン濃度は急激に増加し,その後の減少は他群と比較して遅く,他の3群(12L群,3L群,対照群)が360分経過後には基礎値と有意差がなくなったのに対し,360分経過しても基礎値に戻らなかった。一方,後期分離群(12L群と3L群)の血中コルチコステロン濃度変化には,対照群との間に有意差は認められなかった。早期分離群間の比較では12E群の方が3E群よりも変化の程度が大きく,ピーク後の減少も遅かった。今回の結果から,生後早期(1週齢)に行われた母子分離が,ラットのストレスに対する視床下部-下垂体-副腎皮質系の反応性に長期的影響を与える可能性が示唆された。

In 2018, the group immobility stress was measured in the blood sample. 1. The mother and child segregating groups were divided into 4 groups: (1) early segregating group 12 (12E:6),(2) early segregating group 3 (3E:6),(3) late segregating group 12 (12L:6),(4) late segregating group 3 (3L:6),(5) contrast group 6 (6) and count group 5. 2. Restraint, stress, and body movements The time limit is 30 minutes. 3. Blood sampling was performed after 15 minutes, 30 minutes, 60 minutes, 120 minutes, 180 minutes, 240 minutes, 300 minutes and 360 minutes, and the plasma was frozen and stored remotely. [Results] Early separation group (12E group and 3E group) was reversed, the concentration in blood increased rapidly after the start of restriction, and then decreased. Compared with other groups, three other groups (12L group,3L group, control group) were reversed after 360 minutes, and the concentration in blood increased rapidly after 360 minutes. On the other hand, the blood concentration of the late separation group (12L group and 3L group) changed, and the concentration of the light group changed intentionally. Comparison between early separation groups: 12E group, 3E group, and so on. The results of this study suggest that maternal and child separation during early postnatal period (1 week) may have long-term effects on the hypoblepharo-adrenal cortex system.
