ネスファチン-1の活性部位の同定を試み、中央部分のM30部分のぺプチドのみがネスファチン-1と同様の摂食抑制作用が認められた。M30250 pmol/g体重の単回投与による代謝に及ぼす影響を検討した。雄性ICRマウスにM30を腹腔内投与し、その後1時間の酸素消費量の変化や各種血中代謝マーカーに対する影響について検討を行ったが、明期におけるM30の単回投与実験条件下においてはM30投与による明らかな酸素消費量への影響は認められず、血糖値や脂質濃度にも有意な変動は観察されなかった。また褐色脂肪組織におけるuncoupling protein(UCP)-1の発現にも有意な急性効果は認められなかった。更に予備的な検討として、のネスファチン/ヌクレオビンディン-2遺伝子欠損雄性マウスの酸素消費量の変動に関する検討を行ってみたが、明期中期のみにおいて3時間にわたり測定した酸素消費量に関する検討結果では、明らかな酸素消費へのネスファチン/ヌクレオビンディン-2遺伝子欠損の影響は認められていない。更に自発運動量の計測も予備的に行ってみたが、有意な変動は認められなかった。
