项目摘要

統合失調症疾患感受性遺伝子の中で、最も有望なものの一つにニューレグリン1(NRG1)が挙げられる。2002年の最初の報告以来、多くの追試が行われたが、決定的な結論に至っていない。今回我々は、これまでの追試の中で、最大のサンプル数を用い、人種差を考慮したマーカー選択を行ったうえで、NRG1統合失調症の関連を検討した。包括的に解析を行うため、1)変異検索(5'領域、すべてのエクソン領域)、2)スクリーニング用サンプルを用いた関連解析(1126caseと1022control)、3)追試用サンプルを用いた関連解析(1262case、1172control,166trio sample)を行った。変異検索では、新規の変異は発見できなかった。人種差を反映するべく、NRG1の興味ある領域(HapIce領域とエクソン領域)を代表させる計60個のtagging SNPsを選出し、スクリーニング用サンプルで関連解析を行った。Allele, genotypeとも有意な関連は認められなかったが、一つのhaplotypeで有意差が検出された(P=0.0244,uncorrected)。この有意性を確認するため、追試用のサンプルを用いて解析を行ったが、有意性は追試することができなかった。したがって、当初の有意差は、多重比較によるtype I errorであり、NRG1は日本人統合失調症の疾患感受性に主要な役割を果たしている可能性は低いと考えられた。
