项目摘要

口腔癌において、(1)口腔扁平上皮癌においてRT-PCRによりUPmRNAの発現を確認した。また、細胞株における高発現をWestern blottingにより確認した。(2)口腔扁平上皮癌細胞株を用いてUP高発現癌細胞株の樹立を行った。(3)その増殖能についてコントロール細胞株と比較した所、有意に細胞増殖レベルの上昇が認められた。→UPの過剰発現により癌細胞増殖活性が高まることが示唆された。(4)UP高発現口腔癌株ではp53の発現が低下していることを示す結果が示唆された。(5)口腔癌において、p53を始めとするがん抑制遺伝子、PTHR1、THRBその他3p、9q領域のLocusにおいてLOHおよびMSIが確認された。⇒これにより、UPの過剰発現により癌細胞増殖活性が高まること、UPを介在したp53の発現抑制が、その分子機構として機能している可能性が示唆された。
