エネルギー代謝補酵素(NAD(P)(H))は、生物の様々な代謝過程を制御する重要な補酵素である。本研究では、炭素及び窒素代謝に注目し、NAD(P)(H)による代謝制御機構の解明を試みた。初めに、葉緑体内でNADP を合成する葉緑体局在型NAD リン酸化酵素(NADK2)を高発現させ、NADP 量を増加させた形質転換体を解析した。代謝解析の結果、高発現体ではカルビンサイクルに属する代謝物がルビスコ活性と共に増加していた。さらに、グルタミンとグルタミン酸の蓄積量及び合成速度が増加しており、窒素代謝も促進されていることが明らかになった。また十分な窒素条件下では、野生型よりも良い生長が観察された。本高発現体では総NAD(P)(H)量と酸化還元比は変化していないことから、NADP の絶対量が植物代謝の制御に重要であることが示唆された。
