Es sollen die Kristallisationsbedingungen für Komplexe von Aktin mit Cofilin bzw. ADF verbessert und weiter ausgearbeitet werden. Weiterhin soll der Einfluss von Cofilin und ADF auf das Aktin-Treadmilling in An- und Abwesenheit weiterer Aktin-bindender Proteine wie Thymosin b4, Profilin, Gelsolin, Arp2/3 Komplex und dem humanen AiP1 untersucht werden. Mit kinetischen Methoden sollen die F-Aktin fragmentierenden und die Geschwindigkeit der Dissoziation von Aktinmolekülen vom (-)Ende beschleunigenden Wirkungen von ADF und Cofilin getrennt untersucht werden. Aus Kulturzellen sollen zytoplasmtische bzw. nukleäre sog. Cofilin/ADF Stäbchen isoliert und strukturell und biochemisch untersucht werden.
