Analysis of the mechanism of Calcium-sensing receptor activation in cementoblasts

我们发现，细胞外钙离子水平升高会增加成牙骨质细胞中成纤维细胞生长因子-2基因的表达。蛋白激酶A(PKA)抑制剂H89或腺苷环化酶抑制剂MDL-12330A可抑制钙刺激的成纤维细胞生长因子-2的表达。而蛋白激酶C(PKC)抑制剂GF-109203X或磷脂酶C(PLC)抑制剂U73122均不影响成纤维细胞生长因子-2转录本的表达，提示成纤维细胞生长因子-2的表达增加依赖于蛋白激酶A，而不依赖于PLC/PKC信号通路。

We found that elevated levels of extracellular Ca^<2+> increase fibroblast growth factor (FGF)-2 gene expression in cementoblasts. Pretreatment with a protein kinase A (PKA) inhibitor, H89, or an adenylate cyclase inhibitor, MDL-12330A, inhibited Ca^<2+>-stimulated Fgf-2 expression. In contrast, pretreatment with the protein kinase C (PKC) inhibitor GF-109203X or the phospholipase C (PLC) inhibitor U73122 did not affect the expression of Fgf-2 transcripts, suggesting that the increase in Fgf-2 expression was dependent on the PKA but not the PLC/PKC signaling pathway.
