In der eukaryontischen Zelle wird der Inhalt des Zellkerns und des Zytoplasmas durch die Kernmembran getrennt, welche eine Vielzahl von Kernporenkomplexen enthält. Die Kernporenkomplexe, bestehend aus einer großen Zahl von Untereinheiten, müssen einerseits einen effizienten Austausch zwischen Zellkern und Zytoplasma gewährleisten, andererseits muss der Transport durch sie hoch spezifisch sein. Zur Zeit ist noch völlig unklar welcher Mechanismus für diesen spezifischen aber gleichzeitig effizienten Transport verantwortlich sein könnte. Die vorhandenen Modelle erklären nur Teilaspekte der beobachteten Transportcharakteristik. Im Rahmen des vorliegenden Antrags sollen konformationelle Änderungen des Kernporenkomplexes an intakten aber immobilisierten Kernmembranen mit Hilfe von mechanischen Einzelmolekülexperimenten und der Abbildung der Komplexe mittels der thermischen Bewegung kollidaler Partikel untersucht werden. Speziell sollen mit dem Transport verbundene konformationelle Fluktuationen der Porenkomplexe mit einer mikrosekundengenauen zeitlichen Auflösung erfasst werden. Diese experimentelle Bandbreite kann zur Zeit mit keiner anderen etablierten Technik erzielt werden. Die quantitativen Daten werden uns erlauben bestimmte Transportmodelle auszuschließen. Gegebenenfalls sollen neue Modelle, gestützt durch Computersimulationen, vorgeschlagen werden. Summary: In eukaryotic cells, the nuclear interior and the cytoplasm are separated by the nuclear envelope, which is perforated by large proteinaceous assemblies - nuclear pore complexes (NPCs). Through the NPCs, material has to be efficiently transported between both compartments, but at the same time the transport has to be specific to maintain the separation of nuclear- and cytoplasmic components. The mechanism underlying the transport machinery is currently under strong debate and there are several possible models, which can explain aspects of the experimentally observed transport characteristics. In this proposal, we plan to investigate nuclear translocation mechanisms on immobilised intact nuclear membranes by using single molecule mechanical experiments and three-dimensional thermal noise imaging of single unfixed NPCs. We particularly aim at the characterisation of the conformational dynamics over a wide bandwidth of various parts of the nuclear pore complex, which are not accessible by any other established technique. The gained quantitative information will allow us to distinguish between current translocation models or, potentially, propose a completely new model.
