CD133 have been used to characterize cancer stem cells. In thisstudy, I investigate CD133-positive (CD133+) status in OSCC cell line and molecular biological features. CD133+ has an ability to form sphere in serum-free suspension culture but not CD133- cells. But Sphere formation ability was acquired by adding the a little positive cells. I investigate the expression kinetics of CD133 in sphere formation cells. Sphere has at least one CD133+ cell among CD133- cells and transition of CD133+ to CD133- each other. I compare effect of various growth factors on sphere formation from parental cells, and CD133 expression by flow cytometric analysis. EGF and SHH promote sphere formation, CD133 expression. In addition, CD133+ cells exhibited highly tumorigenic phenotype in nude mouse xenograft. These results suggest that the CD133+ OSCC cells may function as CSCs.
