Structural and functional diversities of mammalian cilia

1)研究了哺乳动物细胞纤毛数量调控的分子机制。基于Transciptome分析和随后的基因敲除实验，我们鉴定了一个新的感兴趣基因。我们构建了CRISPR/Cas9载体，构建了该基因的敲除小鼠。2)为了监测多发性髂骨发生的细胞事件，我们产生了慢病毒载体，用荧光蛋白标记睫状体和基底体。3)研究了胎儿期脉络丛上皮细胞纤毛功能的独特变化。4)分析了TRPV4在CPECs中的纤毛定位及其功能意义。

1) We have investigated the molecular mechanism of ciliary number regulation in mammalian cells. Based on transciptome analysis and the following knockdown experiments, we identified one novel gene of interest. We have constructed CRISPR/Cas9 plasmid to generate knockout mouse of this gene. 2) To monitor the cellular event of multiciliogenesis, we have generated lentiviral constructs to label the ciliary shaft and the basal body by fluorescent proteins. 3) We have investigated the unique functional transition of cilia in choroid plexus epithelial cells (CPECs) from the prenatal period. 4) We have analyzed the ciliary localization and functional significance of TRPV4 in CPECs.
