全身性エリテマトーデス（SLE）患者の血清中に出現する抗リボソームP抗体（抗P抗体）は，中枢神経障害やループス腎炎発症と関連する注目すべき抗体である。既に，研究代表者らは抗Pモノクローナル抗体（抗P抗体m）を作成し，マウスに投与すると不安行動・うつ行動を呈し，その病態は血清トリプトファン濃度低下と関連するとともに抗P抗体陽性SLE患者では血清トリプトファン濃度が低いこと，抗DNA抗体との併存はループス腎炎組織所見の重症化に関与することを明らかにしてきた。本研究は抗P抗体mを有力な研究手段として用いることで，SLE患者における末梢血単核球の抗P抗体エピトープの発現を明らかにし，トリプトファン代謝と炎症性サイトカイン，精神状態やSLE活動性との関連について解析することで，SLE患者における抗P抗体特異的な免疫学的異常の機序と病態形成の解明を行うことを目的とするものである。まず抗P抗体mの反応性を腫瘍由来の細胞株を用いてフローサイトメトリー法で解析したところ、抗P抗体mは細胞表面に表出しているP抗原に反応することがわかった。さらに、健常人の末梢血単核細胞における反応性も確認している。今後SLE患者において検討を進める予定である。また、SLE患者の血清を用いた検討では、抗P抗体価とSLEDAI、炎症性サイトカイン、トリプトファン代謝産物であるキヌレニンとトリプトファンの比（K/T比）の関連について検討中である。
